The two conditions, while seemingly distinct, share significant pathophysiological links that can negatively impact intrauterine growth and, subsequently, the child's long-term health. Understanding the mechanisms underlying inflammation in both hemibodies – maternal (placenta) and fetal (gut) – is fundamental to the development of effective therapeutic strategies. The placenta, as a vital organ for fetal nutrition and oxygenation, can have its function compromised when inflamed, leading to fetal growth restriction. Similarly, fetal intestinal inflammation, often associated with environmental or genetic factors, can alter the absorption of essential nutrients and compromise the integrity of the intestinal barrier, with direct repercussions on fetal development.

A placental inflammation Placental inflammation can be triggered by a variety of factors, including maternal infections, autoimmune diseases, and conditions such as pre-eclampsia. These aggressions can lead to changes in vascularization, increased oxidative stress, and the production of pro-inflammatory cytokines, compromising the exchange of oxygen and nutrients between mother and fetus. This placental dysfunction is a major contributor to intrauterine growth restriction (IUGR), a condition associated with an increased risk of perinatal complications and health problems in adulthood, such as cardiovascular and metabolic diseases. Early identification and intervention in cases of placental inflammation are therefore of utmost importance.

On the other hand, the fetal intestinal inflammation This can result from a complex interaction between the maternal and fetal microbiota, diet, and the fetus's immune response. Dysbiosis during pregnancy, for example, can translocate bacteria or their products to the fetus, triggering an inflammatory response in its gastrointestinal tract. This inflammation can not only affect nutrient absorption but also potentiate systemic inflammation, perpetuating a vicious cycle that impairs fetal growth and development. Research in this area aims not only to understand the mechanisms but also to explore preventive and therapeutic strategies, such as microbiota modulation or the use of specific anti-inflammatory agents.

The synergy between placental and intestinal inflammation is a field that demands in-depth investigation. By addressing these two aspects together, we can open new doors for the development of innovative interventions that promote healthy fetal growth and reduce the incidence of long-term health complications in Cape Verde and globally. Our proposal aims precisely to explore this interconnection, with the goal of developing intervention approaches that can be applied in clinical and public health contexts, particularly in populations with greater vulnerability.

Applications for funding for this project are open until April 28, 2026.

For more information and to submit proposals, please visit: https://gcgh.grandchallenges.org/challenge/novel-interventions-targeting-placental-and-gut-inflammation-improve-fetal-growth

Did you know that inflammation in the placenta and the baby's gut can be linked and affect its safe growth?
